There is a quick and easy way to zoom the video playback window in iTunes to size. This works when you are playing a video in a separate window.

Here’s how:
Control + click (or Right Click for you multi-button mouse users) on the video and choose a zoom level from the pop-up contextual menu.

You can also drag the window to a different size by dragging the lower right window corner.
